Determination of Total Phenolic and Flavonoid Levels of Mangkokan Leaf Extract (Polyscias scutellaria)

Abstract

The fresh leaves of the mangkokan plant contain several chemicals and properties that have bioactivity. This study aims to determine the chemical content, total flavonoid, and phenolic levels in the ethanol extract of mangkokan leaves (Polyscias scutellaria). Extraction was done by the maceration method using 96% ethanol. Determination of phenolic using FeCl3 as a reagent, the blackish-green color indicates the presence of phenol compounds. Flavonoid identification was carried out with reagents between magnesium and concentrated HCl. The existence of flavonoids will give the characteristic orange color. The extract was analyzed by colorimetric method using UV-Vis spectrophotometer. The results showed that the total phenolic and flavonoid content in the ethanol extract of mangkokan leaves was 14.67 mgGAE/g and 1.83 mgQE/g respectively.
